Our recommended workflow

Import calcium movies recorded using nVista, along with metadata and general-purpose input/output traces. Pre-process to remove acquisition-specific artifacts and reduce data size.
Eliminate motion artifacts, which can occur due to motion of the brain relative to the skull. Align individual movie frames for further processing.
Perform spatial normalization so that signals at different locations within the movie are comparable in intensity.
Automatically identify the hundreds of cells located within your movie, and extract the traces associated with them. Manual selection and quality control of cells based on their images and traces is recommended.
Separate cell activity events from noise, and export your traces to simple text format for further analysis outside of Mosaic.
